Investigating VEC behavior is important for understanding the detailed mechanisms of new capillary formation. VEC behavior is influenced by the properties of the extracellular matrix (ECM), such as stiffness and degradation. Hyaluronic acid (HA) is a key component of the ECM that governs VEC behavior through the interaction of CD44 receptors on VEC surface. To investigate these properties in vitro, this study uses hydrogels with tunable mechanochemical properties, made from HA modified with phenol moieties (HA-Ph). These hydrogels are prepared through horseradish peroxidase (HRP)-mediated crosslinking and photocrosslinking, providing a versatile platform to study the effects of hydrogel stiffness and HA-Ph degradation on VEC functions. This seminar introduces the design and application of these HA-Ph hydrogels and discusses how their microenvironmental properties influence VEC adhesion, elongation, and capillary-like network formation.
